Unsere Forschung

Die Arbeitsgruppe von Alexander Westermann erforscht RNA-basierte Mechanismen, die dem Zusammenspiel zwischen Wirt, Krankheitserregern und der natürlichen Darmflora des Wirts zugrunde liegen. Das Ziel ist es, Moleküle und Stoffwechselwege zu identifizieren, die in der Diagnostik und Therapie von Darminfektionen eingesetzt werden können.

Die aktuelle Antibiotika-Krise macht deutlich, wie dringend der Bedarf an alternativen Behandlungsmöglichkeiten für Infektionskrankheiten ist. RNA-basierte Therapien bieten großes Potenzial für die gezielte Bekämpfung von Darmkeimen bei gleichzeitiger Minimierung von Kollateralschäden an der umgebenden natürlichen Darmflora (Mikrobiota).

Alexander Westmanns Arbeitsgruppe erforscht das komplexe Netzwerk von Interaktionen zwischen humanen Darmmikroben, dem Menschen als Wirt und eindringenden Krankheitserregern während Infektionsprozessen. Ihre Forschung zielt darauf ab, sogenannte nicht-kodierende RNA-Moleküle (ncRNA) und RNA-bindende Proteine im Erreger, dem Wirt und seiner Mikrobiota zu identifizieren und ihre Funktion zu entschlüsseln, um die Entwicklung maßgeschneiderter RNA-basierter Diagnostika und Therapeutika voranzutreiben.

Der aktuelle Schwerpunkt der Arbeitsgruppe liegt auf dem Transkriptom des menschlichen Darmkommensalen Bacteroides thetaiotaomicron und seiner Besiedelung des menschlichen Dickdarms. Dazu entwickeln die Forscher derzeit ein hochkomplexes In-Vitro-Modell des menschlichen Darms aus Organoiden und nutzen RNA-Sequenzierungstechnologien, wie Dual RNA-seq und differentielle RNA-seq. Das übergeordnete Ziel des Teams ist es, neue Zielstrukturen für die Behandlung und Prävention von Darminfektionen zu finden, die auf RNA-Mechanismen basieren.

Forschungsprojekte

Bakterielle Infektionen höherer Säugetiere sind hoch komplexe biologischen Prozesse, die in der Regel die Interaktion einer Vielzahl verschiedener Organismen umfassen. Wie können bakterielle Krankheitserreger (Pathogene) eine Infektion im Wirt vorantreiben und welche Abwehrmechanismen müssen sie überwinden, um sich anzusiedeln? Welche molekularen Mechanismen vermitteln die Schutzfunktion der Mikrobiota gegenüber einem eindringenden Erreger? Welche Rolle spielen nicht-kodierende RNAs (ncRNAs) im Zusammenspiel von Wirt, Mikrobiota und Pathogen? Diesen und weiteren Fragen widmet sich unsere Arbeitsgruppe.

Unser Forschungsschwerpunkt liegt auf der Identifizierung und funktionellen Charakterisierung von ncRNAs im Darm-Pathogen Salmonella Typhimurium, dem bedeutenden Mikrobiota-Bakterium Bacteroides thetaiotamicron und dem menschlichen Wirt. Wir verwenden modernste RNA-Sequenzierungs- (RNA-seq) Techniken um gezielt RNAs ausfindig zu machen, die zukünftig als Biomarker in Diagnose und Therapie eingesetzt werden können.

Neben der Entwicklung neuer RNA-seq-basierter Technologien für komplexe Infektionssysteme ist es unser Ziel, das Wissen über die Funktionen von nicht-kodierenden, regulatorischen RNA-Molekülen und RNA-bindenden Proteinen in der bakteriellen Krankheitsentstehung (Pathogenese) und Symbiose zu erweitern, indem wir biologische Erkenntnisse aus mechanistischen Studien gewinnen.
